The Effects of Lithium on Long-Term Liquid Culture System in Murine Hemstopoiesis


Abstract
The role of lithium in increasing granulocyte and platelet products has been studied.
So the authors have studied the stimulation effect on granulopoiesis and
megakaryopoiesis together or singly to see whether it has a direct effect on the stem
cell or an indirect colony stimulating effect where the integrity of T-lymphocycle. And
also to see how were changed the number of colonies during 4 weeks culture used by
liquid system in the murine hematopoietic cells.
The results were as follows :
1. The total colonies cultured to the three weeks was increased 216 percentage
compared with those of the control group and was sustained well until four weeks, but
there was not changed on the second week.
2. With a 2.0 and 4.0 mEq/1 concentration of the lithium cultured in a conditioned
medium and stimulated by lithium alone, those colonies decreased compared with those
of the control. Especially the colonies was lower produced at the 4.0 mEq/1 than those
of 2.0 mEq/1 of concentration due to toxic effect.
3. When the pokeweedmitogen was added to the lithium in order to see the colony
stimulating activity, the total colonies shows increment on 1st & 2nd week.
It was also maintained until on the 4th week in liquid culture system. But there was
no difference in any various concentration of lithium.
4. When cyclosporin 4, an inhibitor of the T-lymphocyte function, was added to the
conditioned medium once with pokeweedmitogen present or once without
pokeweedmitogen, the total colonies were decreased than those of the control group. The
colonies was not changed between with PWM to without PWM added to the 2.0 mEq/1
concentration of the lithium during 4 weeks culture.
The above data suggest that the lithium-induced granulopoiesis and megakaryopoiesis
do not directly stimulate to the stem cell, but cause to an enhancement of colony
stimulating activity where the integrity of T-lymphpocytes is required.
And, so the therapeutic dose of lithium were maintained well until four weeks used
by liquid culture system that would have attempted to treat with lithium in some
patients with cancer induced leukopenia or aplastic anemia.
